一个来自哈佛大学的研究小组近日在国际学术期刊PNAS上发表文章,他们利用延时拍摄的受激拉曼散射显微镜技术观察到了体内细胞分裂过程中DNA的动态变化,并且这种技术不需要对DNA进行荧光标记。

为了深入了解癌症等疾病发生过程中细胞内各种生物过程所发生的变化,就需要在不干扰细胞生命活动的情况下对细胞内部进行细致观察。通常情况下,如果想要对染色体活动进行观察就需要对DNA进行标记,而这种方法具有侵入性可能会改变细胞内的一些天然微环境。

受激拉曼散射显微技术是一种可以观察到化学键振动频率的成像技术,基于所处的分子环境,不同类型的化学键有不同的振动频率。与传统的拉曼光谱技术不同,受激拉曼散射显微技术能够在体内研究中实时地从样品上快速获得数据。利用这种技术,研究人员可以对细胞内DNA,蛋白质以及脂质的丰度和分布进行定位,使得他们可以通过无标记的方式对细胞分裂过程进行观察。

研究人员首先在Hela细胞中观察到了有丝分裂的不同时期,并对DNA,蛋白质以及脂质在细胞内的分布进行了三维重建。随后他们又对促细胞分裂化学剂TPA处理小鼠的皮肤进行了观察,看到了细胞周期的每一个阶段,除此之外,研究人员还利用人类癌细胞处理免疫缺陷小鼠诱导其产生肿瘤,并对癌细胞的染色体运动进行了观察。

这项研究表明无标记受激拉曼散射显微技术可以在自然背景下对细胞内生物过程进行观察,并为追踪细胞行为提供了一种全新的方法,对于癌症等疾病的细胞学研究具有一定意义。
