Cell：瘦素和胰岛素作用于POMC神经元促进白色脂肪棕色化

近日，来自澳大利亚莫纳什大学的Tony Tiganis在国际顶尖期刊cell发表了他们关于瘦素和胰岛素作用于POMC神经元促进白色脂肪棕色化的相关研究成果。文章首次证明瘦素和胰岛素能够协同作用于POMC神经元驱动白色脂肪棕色化以维持机体能量平衡。
 
研究人员指出，瘦素是由脂肪细胞分泌的一种重要的脂肪因子，对能量平衡和体重控制具有重要作用，瘦素能够作用于POMC神经元和NPY/AgRP神经元以抑制食欲促进能量消耗；胰岛素由β胰岛细胞分泌的一种重要因子，能够作用于肝脏、脂肪、肌肉等多种器官组织以调节血糖水平，同时也能作用于POMC神经元和AgRP/NPY神经元，但以往的研究观点认为瘦素和胰岛素作用于不同的POMC神经元亚群以发挥调节能量平衡的作用。该研究证明POMC神经元可以整合瘦素和胰岛素信号，促进白色脂肪棕色化维持能量平衡。
 
该研究利用POMC神经元中PTP1B,TCPTP敲除小鼠和PTP1B，TCPTP双敲除小鼠证明PTP1B和TCPTP选择性调控瘦素和胰岛素信号通路影响了体重,能量消耗和外周血糖平衡；并且PTP1B和TCPTP同时失活，或者在POMC神经元中促进瘦素和胰岛素信号能够增加白色脂肪棕色化以及机体能量消耗，抵抗饮食诱导的肥胖发生。
 
综上所述，该项研究发现瘦素和胰岛素能够协同作用于下丘脑的POMC神经元，可能在一个长期的过程中，使更多POMC神经元参与到白色脂肪棕色化过程，通过增加能量消耗以限制体重增加。
