PNAS:瘦素之外的脂肪稳态新机制

大家都会注意到,每天花费很长时间来静坐的人群肥胖的风险一般会有所增加,但是至今为止,关于减肥效果的机制尚不清楚。瘦素系统是现在唯一已知的调节脂肪稳态的体系。来自哥德堡大学的研究人员假设,除了瘦素,还有另一个调节体重和脂肪量稳态的调节系统。

研究人员通过实验证明,增加啮齿动物的负重,即在其腹部或背部皮下植入不同重量的胶囊,可以通过减少动物食物摄入量,可逆地降低生物体重。重要的是,负重能够缓解饮食诱导的肥胖,并改善葡萄糖耐受。

而且,研究人员此次确定的体重稳态调节是独立于来源于脂肪的瘦素系统的,揭示了用于脂肪量调节的两个独立的负反馈系统。已知骨细胞可以感知骨应变的变化。在这项研究中,负载增加的体重减轻效应在骨细胞耗尽的小鼠中丧失。因此,研究人员认为,增加体重会激活依赖于承重骨的骨细胞传感器。这诱导了传入信号,以减少体重。

这些研究结果揭示了一个独立于瘦素的调节脂肪量的体重稳态系统(“gravitostat”)。
