为了研究EC特异性的转录组基础,Paik等分析了来自Tabra Muris集团的小鼠组织特异性EC的单细胞RNA测序数据。

12种器官内皮细胞的单细胞转录组

经分析发现,通过单细胞RNA测序和组织块RNA测序产生的组织特异性EC转录组数据之间有很强的相关性,验证了该方法的可靠性。采用基于GRAPH的聚类算法,研究人员发现某些组织特定的EC明显根据器官而聚类(如,肝、脑),而其他组织来源的EC的转录组有明显的重叠(如,脂肪、心)。

此外,研究人员还明确了可能与维持其特征的组织特异性EC和信号通路的新型标记。性别是内皮细胞转录组中异质性的重要来源,研究人员发现Lars2基因在雄性小鼠EC中高度富集。小鼠心脏和肺EC的标志物在人类胎儿心脏和肺EC中保守。通过分析配体和受体表达模式,研究人员还确定了组织特异性EC和其他细胞类型之间潜在的血管分泌相互作用。

总之,Paik等人通过对单细胞RNA测序数据进行分析揭示了维持组织特异性EC特征的转录网络,并发现了组织特异性EC之间的血管分泌和功能性相互作用。
