PNAS:放疗变身“癌症疫苗”联合中性粒细胞增强抗肿瘤免疫

放射治疗是三种癌症核心疗法中的一种。研究人员发现靶向肿瘤的放射治疗能够作为“癌症疫苗” 引起中性粒细胞介导的肿瘤细胞死亡,激发免疫系统对抗其他转移灶的癌细胞。

这也叫做“远端效应”,即放射治疗针对原位的癌细胞,随后导致未接受放射治疗的其他转移灶发生萎缩或癌细胞的清除,科学家们在几十年前就已经观察到这一现象。

“远端效应只是偶尔能够观察到,但是一旦出现就会在病人体内诱导长效的抗肿瘤应答。我们进行这项研究希望能够深入理解这一现象,同时找到增强这种效应的方法。”Raquibul Hannan教授这样说道。

研究结果表明,中性粒细胞是在放射诱导的抗肿瘤免疫应答中扮演关键角色。不进行放疗的情况下,癌细胞将中性粒细胞转变成肿瘤相关中性粒细胞帮助促进癌细胞生长。放射治疗不仅摧毁了肿瘤相关中性粒细胞,还会招募新的中性粒细胞进入肿瘤。放射诱导的中性粒细胞(RT-N)会通过产生损伤肿瘤细胞的细胞因子攻击肿瘤细胞。该研究还进一步证明放射诱导的中性粒细胞对于产生下游肿瘤特异性,T细胞介导的抗肿瘤免疫应答有非常关键的作用。

除此之外,研究人员还发现通过给予天然细胞因子G-CSF(粒细胞克隆刺激因子)可以增强放射诱导的中性粒细胞的肿瘤杀伤能力,这种细胞因子能够刺激骨髓产生更多白细胞,其中包括中性粒细胞。在临床上,这种细胞因子广泛应用于因化疗导致的白细胞缺少。研究人员发现G-CSF与放射诱导的中粒细胞的结合能够促进抗肿瘤免疫应答。

“这些结果为放疗与G-CSF在临床前和临床研究中的联合使用提供了支持,”Hannan教授这样说道。“我们的长期目标是希望远端效应能够每次都能够得到诱导发生,而不仅仅是零星出现。”
