Cell Stem Cell:北大学者首次实现了单细胞水平的5-醛基胞嘧啶全基因组测序

来自北京大学生命科学学院,BIOPIC中心的研究人员发表了题为“Single-Cell 5-Formylcytosine Landscapes of Mammalian Early Embryos and ESCs at Single-Base Resolution”的文章,在国际上首次实现了单细胞水平的5-醛基胞嘧啶全基因组测序,并解析了小鼠着床前胚胎中的单碱基分辨率5-醛基胞嘧啶(5fC)谱图。

这一研究成果公布在Cell Stem Cell杂志上,文章的通讯作者分别是北京大学生命科学学院伊成器研究员、汤富酬研究员,第一作者为博士生朱晨旭、高云,郭红山博士以及夏波。

哺乳动物早期胚胎发育过程中会发生剧烈的表观遗传重编程。在5-甲基胞嘧啶的主动去甲基化过程中会产生一种新颖的表观遗传学修饰5fC。

伊成器课题组与汤富酬课题组紧密合作,开发了一种基于特异性化学标记的单细胞、单碱基分辨率的5fC测序技术(CLEVER-seq),并将其应用于小鼠着床前胚胎以及哺乳动物多能干细胞的5fC检测。研究发现,在小鼠早期胚胎发育过程中,5fC发生高度动态变化并富集在基因表达调控区域。5fC在不同的胚胎发育阶段以及多能干细胞中具有高度的异质性。

同时,启动子区域5fC的产生先于相应基因的表达,说明在哺乳动物早期胚胎发育过程中主动去甲基化可能参与了关键基因的表达调控。

汤富酬研究员曾发表综述,深入浅出的介绍了干细胞生物学研究中的单细胞测序技术。单细胞基因组、表观基因组、转录组测序技术最近发展得很快。这些方法被广泛用于不同的干细胞研究,促成了许多激动人心的新发现。汤富酬和文路的这篇文章以表格形式对各种单细胞组学测序技术进行了总结,着重介绍了这些技术在干细胞领域(包括多能干细胞和组织特异性干细胞)的应用。文章末尾还简单探讨了单细胞测序在干细胞领域的未来前景。

干细胞能够分化成为机体内任何类型的细胞,既是研究人体早期发育的理想工具,也是细胞治疗的宝贵资源。细胞间差异和异质性是干细胞群体固有的基本特性。如果我们对一群细胞进行组学分析,这些关键的信息就会被掩盖。单细胞测序技术是近几年备受关注的强大工具,可以在同种干细胞中全面剖析细胞异质性,鉴定不同表型的细胞。

同时,今年伊成器研究员受邀发表了题为“Epitranscriptome sequencing technologies: decoding RNA modifications”的综述,介绍了真核生物mRNA上已知的几种主要修饰的检测技术,并对目前表观转录组分析技术存在的挑战及未来的发展方向进行了展望。

在这篇综述中,研究人员对于真核生物mRNA上已知的几种主要修饰(包括N6-methyladenosine,N6,2’-O-dimethyladenosine, 5-methylcytidine, 5-hydroxylmethylcytidine, inosine, pseudouridine andN1-methyladenosine)的检测技术进行了综述,探讨了用于描述这些表观转录组标记的测序技术,比如规模,分辨率,定量特征,预富集能力和相应的生物信息学工具。同时也针对不同检测技术和RNA生物学新方法进行了选择指导,最后对目前表观转录组分析技术存在的挑战及未来的发展方向进行了展望。

Stem Cells:核内肌动蛋白的结构可以调节间充质干细胞的分化

肌动蛋白的结构有助于细胞核内的生理过程以控制间充质基质细胞(MSC)的分化。连续的细胞松弛素D(Cyto D)可以破坏MSC肌动蛋白的细胞骨架从而导致成骨或脂肪的分化形成,这些过程均需要将肌动蛋白转移到细胞核中。Cyto D存在细胞质中,因此核内肌动蛋白的聚合通过肌动蛋白的转移增强。近期一项发表在杂志Stgem Cells上的研究评估了肌动蛋白的结构是否影响分化。此项研究通过成骨和脂肪形成的生物标
