此项研究通过成骨和脂肪形成的生物标志物表达谱和无偏差的RNA-seq分析显示,Arp2 / 3复合体分支的次级肌动蛋白丝是成骨所必需的,并且防止肌动蛋白分支刺激脂肪的形成。

Cyto D可以激活成骨细胞主调节子(例如Runx2,Sp7,Dlx5)和新的配对基因(例如Atoh8,Nr4a3,Slfn5)。

Formin诱导的主要肌动蛋白丝形成对于Arp2 / 3复合物的募集至关重要:通过沉默成蛋白mDia1,而不是其旁系同源mDia2来防止成骨。

此外,虽然抑制肌动蛋白,支链仍是有效的成脂刺激物,沉默mDia1或mDia2可以阻断脂肪形成基因的表达。

此项研究提出假设,mDia1定位在多潜能MSC的细胞质和细胞骨架破裂后的细胞核中,加入核内mDia2以促进初级细丝形成,然后通过Arp2 / 3复合物的募集介导后续分支。产生的核内支链肌动蛋白网络导致了成骨分化,而在不存在Arp2 / 3复合物介导的次级分支的情况下,肌动蛋白聚合引起脂肪形成分化。
