JCC: 丁酸不能预防溃疡性结肠炎患者上皮屏障功能障碍

使用永生化的癌细胞系进行的体外研究表明,丁酸对上皮屏障的完整性具有正向影响,因此,本项研究旨在从溃疡性结肠炎[UC]患者的人体组织样本评估丁酸盐对上皮屏障功能的影响。

背景和目标
使用永生化的癌细胞系进行的体外研究表明,丁酸对上皮屏障的完整性具有正向影响,因此,本项研究旨在从溃疡性结肠炎[UC]患者的人体组织样本评估丁酸盐对上皮屏障功能的影响。

方法
研究人员从UC患者[ n = 10]和非UC对照[ n = 10]的原代上皮细胞中建立单层的上皮细胞体系。用8mM丁酸钠±肿瘤坏死因子α[TNFα]和II型干扰素[IFNγ]处理单层肠上皮细胞48小时。监测跨上皮电阻的变化,并测量屏障基因表达水平。用OLINK定量细胞上清液中的炎症蛋白水平。

结果
本项研究结果显示原代单层培养物可以在培养的1周内以稳定的抗性值和极化紧密连接生长。丁酸盐处理的培养物增加了抗药性,但与TNFα和IFNγ结合使用时有害。联合治疗还进一步诱导了更高的IL8 mRNA和炎性蛋白分泌。在来自患者和非UC对照的培养物中观察到的效果相似。

结论
本项研究表明丁酸盐不能预防炎症引起的屏障功能障碍,甚至会恶化其在UC患者和对照的初级上皮单层细胞中的作用。
