西班牙贝拉特拉大学动物生物技术与基因治疗中心Ruberte J教授及其工作团队近日在Invest Ophthalmol Vis Sci.上发表了他们的最新成果，发现细胞衰老与人类视网膜微动脉瘤形成相关。

微动脉瘤常常发生在健康老年人视网膜中。然而到目前为止，没有研究结果表明与年龄相关的致病机制参与其形成。作者们分析了老年人视网膜和老龄小鼠视网膜中的细胞衰老情况、老化的特征以及几种年龄相关的疾病。

作者们从17名非糖尿病供体和Bmi1缺陷的小鼠获得视网膜，并通过免疫组织化学分析、衰老相关β-半乳糖苷酶活性测定、苏丹黑B染色、常规透射电子显微镜和免疫电子显微镜等方法分析细胞衰老情况。

研究结果表明，神经元和血管在老年人视网膜中执行细胞衰老过程。经典衰老标记物p16、p53和p21上调并与老年人类微动脉瘤细胞凋亡共存。衰老内皮细胞不连续地被纤连蛋白覆盖，p16与纤连蛋白受体α5β1整联蛋白的β1亚基在内皮细胞膜下共定位，这些提示缺氧作为参与内皮细胞凋亡的机制。在缺乏Bmi1的老龄小鼠模型中，p21被过度表达，视网膜血管显示出以穴状窝肿和脂褐素积聚为特征的老化表型。虽然小鼠视网膜不易发展微动脉瘤，但Bmi1缺陷小鼠呈现丰富的视网膜微动脉瘤。这些结果揭示了细胞衰老在老年人视网膜中形成微型动脉瘤过程中的角色定位。
