PNAS:为何你不能代谢掉草酸盐?

然而有些植物、真菌和细菌却可以破碎草酸盐分子,如今刊登在国际杂志PNAS上的一项研究论文中,来自MIT等处的科学家们通过研究鉴别出了代谢草酸盐过程中的一种特殊的分子机制,这对于后期科学家们开发新型方法来减轻或消除草酸盐分子对人类机体的损害提供了新的思路和希望。

研究者Catherine Drennan教授表示,文章中我们利用X射线晶体学技术精确鉴别出了这种名为硫胺素焦磷酸依赖的草酸盐氧化还原酶(OOR)如何利用诱骗法来代谢草酸盐,这种特殊酶类可以轻松分解掉草酸盐;草酸盐分子由两个碳原子和四个氧原子共价结合,碳原子可以彼此互联,每一个碳原子可以单独连接两个氧原子;草酸盐氧化还原酶是一种大型的复杂分子,其可以以特殊的方式结合草酸盐来中和负电荷,同时还会以一种单独的方式来破碎草酸盐中心的碳-碳键,这就是研究人员称之为的诱骗过程。

由于草酸盐氧化还原酶可以利用正电荷吸引负电荷从而结合草酸盐,随后通过改变两种蛋白的运动方式来破碎草酸盐,在OOR和草酸盐反应过程中通过利用一系列的X射线技术,研究人员就可以清楚地观察到OPP代谢草酸盐的动力学过程。

研究人员希望鉴别出草酸盐破碎的机制可以帮助科学家们寻找特殊的方法来帮助个体减少所需的草酸盐的水平或降低草酸盐在机体中积累的水平,有些个体机体中会携带代谢草酸盐的细菌,然而有些个体中却并没有。后期研究人员或将大量开展人类机体中微生物的代谢研究,从而为改善人类健康带来帮助。
