在眼科药物开发和临床前评估期间测试时,通常使用动物进行药代动力学研究。这些测试中的不准确性、成本和道德等因素提示,针对角膜的研究需要构建体外模型。

通过纤连蛋白的方法对膜表面进行功能化处理,测量膜的机械性能和表面性质,以确保可以正确的模拟体内特征。将永生化人角膜上皮细胞培养在膜上,在芯片上创建一个微型角膜上皮(角膜芯片)。构建实验验证其功能,具体是通过使用模型药物,测试人角膜上皮的屏障性能。建立脉动液流模型,可以非常相似地模拟眼前角膜环境,并且模拟体内条件下的渗透性。

因此,他们认为,他们建立的体外角膜模型,可用于潜在治疗药物的临床前评估和模拟人类角膜的环境。
