PNAS:卢煜明教授发表新论文,除了NIPT,游离RNA也有望无创检测了

近日,香港中文大学卢煜明教授领导的团队利用单细胞转录组(single-cell and cell-free plasma RNA transcriptomics)技术,全面解析了人类胎盘的细胞异质性。这项研究以“Integrative single-cell and cell-free plasma RNA transcriptomics elucidates placental cellular dynamics”为题,刊登在近日的《美国科学院院报》(PNAS)上。

早在1997年,卢煜明教授就发现了孕妇外周血中存在游离的胎儿DNA,并发展出了一套新技术来准确分析和度量母亲血浆内的胎儿DNA,即无创DNA产前检测(NIPT)。NIPT作为一种安全、准确、快速检测胎儿重大遗传疾病的新型产前检测技术,可避免传统产前检测技术存在的弊端,目前已应用于全球90多个国家,使数百万孕妇受惠(仅在中国,每年就有一百多万孕妇接受NIPT),大大减少了出生缺陷的发生。

胎盘细胞的异质性是指在不同个体间从基因型到表型上存在的差异。尽管过往研究利用全组织分析鉴定出先兆子痫胎盘表达谱的明显变化,并发现在妊娠和妊娠并发症期间,靶向RNA转录(targeted RNA transcripts)水平增加,母体游离核酸(cell-free nucleic acids)的总量和贡献值也增加。但目前仍然无法通过分析母体游离核酸来无创性检测胎盘细胞动力学和功能异常。



在这项研究中,卢煜明教授领导的团队利用大规模的微流体单细胞转录组技术(large-scale microfluidic single-cell transcriptomic technology),分析了24,000多个非标记细胞(nonmarker selected cells ),确定了细胞类型特异的基因特征,这些细胞来自足月和早期先兆子痫的胎盘。



研究人员利用获得的数据集,鉴定出了人类胎盘中的不同细胞亚型,并重建了滋养层的分化轨迹。通过对母体血浆游离RNA的综合分析,他们分析出怀孕过程中造血细胞和胎盘细胞的纵向发展动力学。

此外,通过血浆游离核酸的分析,研究人员能非侵入性地发现先兆子痫胎盘中绒毛外滋养层细胞的功能异常,这有助于疾病的早期诊断。这项研究表明:来自单细胞的转录组信息或有助于阐释游离血浆RNA,以便在复杂的病理状况下对细胞动态进行非侵入性检测。
