近期一篇发表在Environ Microbiol杂志上的文章,报道了一项独特的体外培养方法,即在没有任何外源性物质(如支架或生长因子)的情况下,在本体组织内培养人3D淋巴网络的生成。

将人体淋巴管内皮细胞(LECs)与皮肤成纤维细胞一起培养,就可以自发地组成了一个稳定的3D毛细淋巴管网络。

该培养方法的第一步,就是将原始的纤维母细胞与LECs从人体皮肤中分离,纤维母细胞经培养后可产生富含细胞外基质的结缔组织(第二阶段);而淋巴管内皮细胞则种植在其表面,以形成一个网络(第三阶段),在液-气界面上出现了组织层的堆积以及组织成熟后(第四阶段),就可以通过显微镜对包含淋巴微血管网络的三维结构进行分析(第五阶段)。

由这种方法培养出来的淋巴管,含有体外培养的人体皮肤淋巴微血管中的主要细胞以及超微结构特征,并且在未来的几个星期里都可以稳定的存在。
