代谢组学分析可以提供揭示与糖尿病病理生理学相关的代谢途径并改善糖尿病风险预测的潜力。研究人员前瞻性地分析了从基线(1987-1989)和事件性糖尿病到20151231日的2939例动脉粥样硬化风险社区(ARIC)研究参与者和无糖尿病患者的代谢组学数据,使用非靶向方法测定他们血清标本中的已知代谢物。

在鉴定的245种已知化合物中,7种代谢物在Bonferroni校正和协变量校正后与事件性糖尿病显著相关; 这些包括食品添加剂(赤藓糖醇)和涉及氨基酸代谢的化合物[异亮氨酸,亮氨酸,缬氨酸,天冬酰胺,3-4-羟基苯基)乳酸酯]和葡萄糖代谢(海藻糖)。更高水平的代谢物与糖尿病发病风险增加有关(异亮氨酸的HR每增加1个标准差2.96,95CI 2.02,4.35p= 3.18×10 -8 ;海藻糖每增加1HRHR 1.16,95CI 1.09 1.25p = 1.87×10 -5),但与糖尿病风险较低相关的天冬酰胺除外(天冬酰胺0.78,95CI 0.71,0.85p = 4.19×10 -8)。这7种代谢物适度地改善了空腹血糖以外的糖尿病事件预测,并建立了危险因素(C统计值0.744 vs 0.735C统计差异p = 0.001)。

支链氨基酸可能在糖尿病发展中起作用。研究人员的研究是首次报道天冬酰胺作为糖尿病风险的保护性生物标志物。血清代谢组反映已知和新型代谢紊乱,并可改善糖尿病的预测。
