Otol Neurotol:羟苯磺酸钙对庆大霉素诱导的耳蜗神经毒性有治疗作用吗?

氨基糖苷类的耳毒性作用是众所周知的。庆大霉素有很大的听力损失风险。尽管庆大霉素有耳毒性作用,但它被广泛用于防治威胁生命的感染。羟苯磺酸钙由于具有血管保护和抗氧化作用,是一种能够用于治疗许多疾病的药理活性物质。最近,有研究人员研究了羟苯磺酸钙对庆大霉素诱导的动物模型耳蜗神经毒性的治疗作用。

研究人员将32只Sprague Dawley大鼠分为四组。庆大霉素、庆大霉素+羟苯磺酸钙、羟苯磺酸钙和对照组。术前和术后的听觉阈值采用听觉脑干反应阈值、点击和16-kHz音波刺激进行测定。研究发现,庆大霉素+羟苯磺酸钙组的点击诱发听力平均比庆大霉素组好27dB(P<0.01);16kHz音波刺激在不同组之间差异不显著(P>0.01)。组织学检查表明对照组和羟苯磺酸钙组的超微结构外观正常,而庆大霉素组表现出最严重的组织学改变,包括髓鞘破坏、总轴突退化和水肿。与庆大霉素组相比,庆大霉素+羟苯磺酸钙组的组织学损害证据明显减少。

最后,研究人员指出,庆大霉素治疗的基础上加用口服羟苯磺酸钙能够对点击诱发的听觉阈值产生有益的影响,组织学研究结果证明了上述结果。
