与年龄相关的线粒体功能障碍和氧化损伤是多种健康问题的主要原因,包括肌肉萎缩和血管疾病(CVD)。尽管Nrf2(一种调节细胞保护基因表达的转录因子)在肌肉相关疾病中的作用仍然不甚明确,但它已显示出对肌肉萎缩和心血管疾病的有益特性。

Sulforaphane(SFN)是一种天然化合物,其可以通过激活Nrf2相关基因,保护细胞功能完整性。SFN已被报导在包括CVD的多种疾病状态中提供保护,从预防癌症到降低胰岛素抵抗。目前,SFN正处于临床试验的不同阶段。

最近,研究人员在Aging Cell发表了一项研究,旨在确定SFN是否可以预防心脏和骨骼肌的年龄相关功能丧失。研究人员使用了2个月大和21~22个月大的小鼠群组,给予常规啮齿动物饮食或补充SFN的饮食12周。在研究完成时,研究人员测量了小鼠的骨骼肌和心脏功能、线粒体功能和Nrf2活性。

该研究显示,与年轻组相比,老龄对照组小鼠的Nrf2活性和线粒体功能显著下降,同时骨骼肌和心脏功能也有所下降。而SFN处理使得老龄小鼠的Nrf2活性、线粒体功能、心脏功能、运动能力、葡萄糖耐量和骨骼肌卫星细胞的活化/分化均得到恢复。

因此,该研究结果表明,年龄相关的Nrf2信号活性的下降和相关的线粒体功能障碍可能与年龄相关疾病过程的发展有关。因此,通过SFN恢复Nrf2活性和内源性细胞保护机制,可能是一种安全有效的策略,以防止因衰老导致的肌肉和心脏功能障碍。
