类风湿关节炎是一种由免疫系统紊乱导致的,以关节炎症为主要特征的风湿病。该病发病率高,致残率高。研究认为,类风湿关节炎65%的发病风险是由遗传因素造成的。此前,科研人员已在整个基因组中找到了59个与类风湿关节炎发病风险相关的基因。

研究中,中方团队开展了大规模的遗传流行病学研究,涉及数千位中国汉族病人,参与找到了类风湿关节炎发病的42个新基因,使该疾病已知的易感基因数目达到了101个,并鉴定了一系列之前未报道的与汉族人类风湿关节炎发病风险相关的基因,同时揭示了与欧洲类风湿关节炎病人在遗传背景中的异同。

该项目中方团队带头人、第二军医大学附属长征医院风湿免疫科主任徐沪济教授说:“这项研究证明,欧洲人的类风湿关节炎和汉族人的类风湿关节炎在遗传学上是比较相似的。因此推知,在一个种族中比较有效的疗法,也会在另一个种族中奏效。”另外,科研人员在汉族人类风湿关节炎研究中发现,该疾病主要有 ACPA阳性和ACPA阴性两种分型,由于二者具有类似的病因,所以一种有效的治疗方法对这两种类型的患者均有效。 此次新发现两个SNP:rs12617656定位于DPP4,rs12379034定位于 CDK5RAP2.徐沪济科研团队主要参与完成的相关研究结果同期独立发表在国际风湿病学杂志《关节炎和风湿病》上。
