蛋白粘斑的聚集发生于很多神经退行性疾病,如老年痴呆症、帕金森氏病和肌萎缩性侧索硬化症(又叫卢伽雷氏病)等。如研究发现老年痴呆症患者会有tau蛋白的累积,这些蛋白在神经细胞中聚集会形成缠结物。这一过程被称为蛋白致病,很多不同类的脑部疾病都存在不同种类蛋白的积累,且在疾病的发生过程中有重要作用。而在本研究中,他们发现一种酶可能具有阻止蛋白致病这一过程的功能。

高水平的NMNAT2可能减缓认知功能的下降

为验证他们的假设,该研究团队分析了500多例老年死者的大脑,这些老年人生前都参与过拉什记忆与衰老项目,在那个项目中,每位参与者的认知功能每年都被评估。研究者们关注烟酰胺单核苷酸腺苷酰转移酶2(简称NMNAT2)的水平,该酶可以生成尼古丁腺嘌呤二核苷酸(NAD),从而保护我们的大脑免受神经细胞活动过多导致的氧化损伤。通过分析,研究者们发现脑部NMNAT2水平较高的成年人认知下降的可能性较低,而该酶水平较低的人更有可能患痴呆症。这一发现表明除了保护大脑免受氧化压力伤害,NMNAT2还可以保护大脑免受记忆和学习能力的损伤。

NMNAT2水平高的小鼠tau蛋白水平低

接下来,该研究小组分析了tau蛋白水平较高的小鼠的大脑,他们发现海马区NMNAT2水平较高的小鼠tau蛋白的水平较低。

此外,他们发现在老年痴呆症患者的脑部,NMNAT2的水平比正常人的少50%,因此,他们推断加强NMNAT2的功能可能为保护认知功能提供有效的治疗方法。总之,研究者们相信这一发现可能为很多神经退行性疾病的治疗提供新的思路和方法。
