Sci Rep:当归补血汤配伍药效物质基础研究取得新进展

上海中医药大学附属龙华医院王佑华副主任医师从香港中文大学、澳门大学访学回院后引进中医药血管内皮功能研究技术方法、中医药斑马鱼药物筛选和机制研究技术方法。周昕主任药师课题组借助龙华医院药物分析平台,在王佑华副主任医师课题组配合下,对不同配伍比例的当归与黄芪进行了系统的药效物质基础的研究。

课题组首先考察了不同比例复方配伍的水煎液指标成分含量,并对其进行综合评分。结果表明:相比于单味药水提物,复方水煎液可有效地促进指标成分如阿魏酸、毛蕊异黄酮等的溶出,且经典复方配伍比例5:1综合评分得分最高。其次,应用斑马鱼模型研究了当归补血汤促血管新生的药效,发现当归补血汤复方、单味药材水煎液能恢复被血管内皮生长因子受体抑制剂诱导损伤的斑马鱼血管,且经典配伍比例的复方水煎液对血管的保护效果强于单味药材水煎液;在复方提取物中,多种指标成分对受损血管的保护能力较强,具有显着促血管新生的作用。对血管内皮功能的研究结果表明:经典配伍比例的当归补血汤复方和黄芪、当归水煎液、以及阿魏酸、毛蕊异黄酮苷等指标成分对溶血磷脂酰胆碱损伤的大鼠胸主动脉血管内皮功能具有显着的改善作用,且复方效果较好。上述结果说明,当归补血汤5:1的经典配伍是最佳比例,是有科学依据的。相比其他比例,无论是有效成分的含量、促血管新生和改善血管内皮功能的效果均较好,且药效物质基础是当归中的阿魏酸、多糖及黄芪中皂苷和黄酮类成分。

这些研究结果为当归补血汤临床应用和促血管新生、改善内皮功能的物质基础研究提供了科学的依据,并于2017年3月22日在国际期刊《scientific reports》(2017 Mar 22; DOI: 10.1038/srep45111, IF=5.228)上在线刊出。
