N6-methyladenosine(m6A)是真核生物mRNA和长非编码RNA上最普遍的一种RNA修饰,介导了超过 80%的RNA碱基甲基化。人们已经陆续鉴定了m6A所需的“读”、“写”和“擦除”蛋白,但对其生物 学功能还知之甚少。

研究人员发现,发育中的果蝇神经系统存在高水平的m6A RNA修饰,而且这种修饰对神经系统的功能 非常关键。干扰生成m6A的分子通路会造成果蝇行为异常,使果蝇不能正确折叠翅膀,无法给自己定 向,移动也更为缓慢。果蝇运动能力受到的这些影响都来自于大脑功能损伤。这项研究还表明,m6A 在调节性别决定的时候也很重要。

“mRNA上带有丰富的RNA修饰,我认为这是近15年来该领域最激动人心的发现。我们揭示了m6A在果 蝇神经系统和性别决定中的重要作用。鉴于这种修饰在脊椎动物神经系统也很丰富,它很可能在人体内也承担了类似的功能,”文章通讯作者Dr. Jean-Yves Roignant说。

这项研究不仅发现了m6A在胚胎发育中起到的作用,还鉴定了m6A甲基转移酶的一个新组分。研究人 员相信,RNA修饰的这个新兴领域(表观转录组)未来还将迎来更多振奋人心的发现。
