研究作者、密歇根大学综合癌症中心主任、著名肿瘤学教授Max S. Wicha博士说:“很早以前我们就知道炎症和癌症之间存在着一些重要的关联,其中包括调控正常和癌症干细胞的一些相似信号通路。”

“这项工作有助于解释在受到损伤后这些信号通路会在正常组织中关闭,而在癌症中仍旧活化导致癌症干细胞增加的原因。此外,他们提出阻断这些炎症环路有可能是靶向癌症干细胞,改善患者预后的一种方法。”

三阴性乳腺癌是一种雌激素受体(ER)、孕激素受体(PR)和人表皮生长因子受体2(Her-2)均为阴性的乳腺癌类型——这三种蛋白是当前治疗的重要靶点。因此目前没有针对三阴性乳腺癌的分子靶向疗法。罹患这种形式疾病的患者往往预后较差。

研究人员在三阴性乳腺癌小鼠模型中测试一种叫做硼替佐米(bortezomib)的药物,发现其可以阻止SOCS3蛋白降解,导致这一炎症环路关闭,减少癌症干细胞,由此阻断了转移。目前硼替佐米已获得批准用于治疗多发性骨髓瘤。

该研究小组以往证实在HER2阳性的乳腺癌中IL6可刺激乳腺癌干细胞,他们现正设计一项使用IL-6阻断剂的临床试验。新研究表明,将硼替佐米添加到IL-6抑制剂中或许是一种靶向三阴性乳腺癌中干细胞的方法。


“现在我们揭示了在三阴性乳腺癌中炎症的调控机制,我们希望可以将我们的研究转化到临床。一些可阻断这些化学信使的药物已被批准用于治疗类风湿关节炎和其他炎症相关基因,这应该可以推动将它们应用于癌症,”研究作者、乔治亚摄政大学癌症中心助理教授Hasan Korkaya博士说。

在开展临床试验之前还需要进行更多的实验室测试。研究人员还认为这一信号通路有可能还适用于其他的癌症,并正对此开展深入的调查。

根据美国癌症协会的统计,今年有235,030名美国人确诊乳腺癌,有40,430将死于这一疾病。
