日本庆应大学医学部的研究团队采用患者身上提取的结肠癌细胞进行培养,再用干细胞特有的 LGR5 遗传因子进行荧光标识,然后移植到小鼠身上,进行治疗实验。结果发现,虽然药物可以杀死干细胞,使癌细胞停止增长,但是停止用药后,干细胞会再次出现,导致癌细胞继续扩大。这是由于从干细胞分化来的短命细胞会发生“返祖”现象,使干细胞重新复活。正常状态下的肠道中,存在着干细胞和短寿命细胞。干细胞在自己增殖的同时会分化

这是由于从干细胞分化来的短命细胞会发生“返祖”现象,使干细胞重新复活。正常状态下的肠道中,存在着干细胞和短寿命细胞。干细胞在自己增殖的同时会分化产生短寿命细胞,而短寿命细胞则不具备增殖能力。这两种细胞由不同的遗传因子控制,即使消灭了导致结肠癌转移和再发可能的肠癌干细胞,从干细胞分化来的短命细胞也会发生“返祖”现象,使干细胞重新复活。

这一结果给根治肠癌解明了大肠癌复发的机理,通过将干细胞治疗与现有药物组合使用,将给根治大肠癌带来希望。
