近日,一项刊登在国际杂志Science Advances上的研究报告中,来自加利福尼亚大学的研究人员通过研究发现了一种简便且有效的方法,即给诱导人类多能干细胞“喂以”机体中的天然分子腺甘酸,就可以使其再生成为骨质组织,这种源于干细胞的骨质组织可以在不产生肿瘤或诱发感染的情况下帮助修复小鼠的颅骨缺陷。

该项研究或将帮助研究者开发治疗严重骨质缺失及遭受外伤性骨创伤患者的新型再生医学疗法,同时研究者或许也能够开发出一种低廉方便的方法制造纯种的成骨细胞。研究者Shyni Varghese教授表示,我们的一个研究目的就是通过开发有效简便的方法来对人类细胞和组织进行工程化操作,从而开发新型的再生疗法,诱导多能干细胞能够转变为机体中任何一种类型的细胞,比如肌肉细胞、心脏细胞等,这个过程称之为分化,正由于具有这种特性,因此诱导多能干细胞才具有一定的治疗潜力,帮助修复或再生多种组织和器官。

然而促使诱导多能干细胞分化成为任何一种类型的细胞并不简单,干细胞的直接分化就好比遵循一种复杂的菜谱一样,其中包含了多种组分和步骤;而科学家们面对的另外一种挑战则是如何制造出不能产生肿瘤的器官或组织(基于干细胞分化产生),研究者通过研究发现,他们可以通过向培养基中添加腺苷酸来控制人类诱导多能干细胞分化成为功能性的成骨细胞,这就好比机体中的活体骨质细胞一样,产生的成骨细胞就可以构建含有血管的骨质组织,当将这些新生的骨质组织移植到骨质缺陷的小鼠机体中时,成骨细胞就可以在不产生任何肿瘤的情况下形成新生的骨组织。

研究者Varghese说道,这让我们觉得非常不可思议,一种单一分子竟然可以指导干细胞的命运,而且我们并不需要制造由小分子、生长因子或其它混合物组成的混合制剂;此前研究中我们通过研究揭示了骨组织中的磷酸钙如何诱导干细胞分化成为成骨组织,而且干细胞可以摄入磷酸钙来产生ATP,随后破碎腺苷酸并且向干细胞发送信号使其转化成为成骨细胞。

如今研究者想知道如果绕过上述途径,仅仅添加腺苷酸会是什么反应,目前他们正在进行更为深入的研究来阐明腺苷酸信号如何促进骨质形成,截至目前为止,他们发现在干细胞表面发现了一种名为A2bR的受体,该受体可以结合腺苷酸,但其中所涉及的机制目前还不清楚。
