该首次证明,对于皮肤鳞状细胞癌来说起源癌细胞控制着EMT过程。相关研究结果发表在国际学术期刊Cell Stem Cell杂志上。

在这项新研究中研究人员利用基因工程小鼠对起源癌细胞进行谱系追踪,并检测表皮不同部位诱导癌症形成的突变基因的表达。令人惊讶的是,起源于毛囊的肿瘤通常都是会发生EMT过程并具有很强侵袭性的间充质样肿瘤,而起源于毛囊间表皮的皮肤癌是上皮高分化的癌症。这些数据表明,毛囊干细胞和它们的后代容易发生EMT过程并发生转移,说明起源癌细胞与肿瘤的侵袭性密切相关。

为了解其中的机制,研究人员对不同起源癌细胞及相对应肿瘤的转录谱和表观遗传谱的差异进行了研究。他们发现,起源癌细胞的表观遗传学和转录图谱会影响致癌基因靶细胞向上皮高度分化型或更具侵袭性方向的转变,这表明起源癌细胞在这种转变过程中发挥重要作用。研究人员表示参与EMT的基因在起源癌细胞中就已经启动,这会促进癌细胞发生EMT过程。

文章作者指出:“结合全基因组转录谱和表观遗传图谱分析,我们现在可以确定哪些基因能够结合特定的DNA区域,调节肿瘤细胞基因表达。发现这些调控不同肿瘤功能的基因网络有助于设计阻断肿瘤发生和EMT过程的新策略,帮助提高肿瘤对治疗的应答,减少转移。”
