J Periodontal Res: :二羟基-2,3-戊二酮及其类似物作为上皮完整性调节因子

群体感应分子调节细菌在生物膜内的行为,同时在宿主组织中引发免疫应答。我们的目的是研究二羟基-2,3-戊二酮(DPD),通用自诱导剂-2(AI-2)及其类似物(乙基-DPD,丁基-DPD和异丁基-DPD)的前体对于牙龈上皮细胞完整性的调节作用。

群体感应分子调节细菌在生物膜内的行为,同时在宿主组织中引发免疫应答。我们的目的是研究二羟基-2,3-戊二酮(DPD),通用自诱导剂-2(AI-2)及其类似物(乙基-DPD,丁基-DPD和异丁基-DPD)的前体对于牙龈上皮细胞完整性的调节作用。

将人牙龈角质形成细胞与四种浓度(10μmol/L,1μmol/L,100nmol/L和10nmol/L)的DPD及其类似物一起温育24小时。使用增殖试剂盒测定活细胞数,通过明胶酶谱法测定基质金属蛋白酶(MMP)-2和-9活性,并分别通过蛋白质印迹法和RT-qPCR测定occludin蛋白和occludin mRNA的表达。

结果:在用100nmol /L丁基-DPD孵育的牙龈角质形成细胞中观察到细胞增殖增加。在用10μmol/L乙基-DPD孵育的细胞中MMP-9活性升高。另一方面,当用/不用DPD或类似物孵育牙龈角质形成细胞时,MMP-2活性未显示任何显着变化。蛋白质印迹分析证明了occludin的五种形式(105,61,52.2,44和37kDa)。用1μmol/L和100nmol/L的DPD和10nmol /L的乙基-DPD孵育增加二聚体(105kDa)形式的occludin,而用100nmol /L的异丁基-DPD孵育增加单体(61 kDa)形式。 DPD和乙基-DPD降低,100nmol /L的异丁基-DPD和10nmol/L的丁基-DPD增加,单体(52.2kDa和44kDa)形式的occludin,而乙基-DPD减少和异丁基 - DPD增加,低分子量(37kDa)形式。根据RT-qPCR分析,牙龈角质形成细胞暴露于10μmol/L的异丁基-DPD上调了occludin的表达。

结论:结果表明异丁基-DPD具有通过刺激闭塞的形成来增强上皮细胞完整性的潜力,而不影响暴露细胞的增殖或明胶酶活性。该研究首次验证了AI-2类似物对上皮细胞反应的调节作用。
