来自美国明尼苏达大学Hormel研究所、德克萨斯大学M.D.安德森癌症中心等机构研究人员证实,己糖激酶Chexokinase-2(HK2)介导的Warburg效应是PTEN- 和p53-缺失驱动前列腺癌细胞生长的必要条件。这一研究发现发表在9月11日的 Cell Reports 杂志上。


领导这一研究的是明尼苏达大学的华人科学家邓义斌(Yibin Deng)教授。其在人类癌症细胞死亡通路、P53对线粒体的调节和TNF对细胞死亡受体的调节等细胞死亡通路方面研究具有巨大成就。近年来,先后以第一作者或者通讯作者在Cell、Nature、PNAS等顶尖一流杂志上发表论文10余篇。

在美国,前列腺癌是仅次于肺癌的第二大男性癌症相关死亡原因。前列腺癌主要是由前列腺上皮内瘤(prostatic intraepithelial neoplasia,PIN)所引起,这种癌前病变最终发展成为了腺癌和转移性癌症。前列腺癌的发生、发展和转移涉及Pten和p53等多个肿瘤抑制基因改变。

在原发性前列腺癌中往往缺失一个PTEN等位基因,在70%的人类晚期前列腺癌中p53常常完全丢失或是发生突变。近期一些全基因组/外显子组测序分析的结果也揭示在致命的去势抵抗性前列腺癌CRPC)中 PTEN和p53常共同删除或是存在共同突变。但到目前为止,PTEN和p53缺失驱动前列腺癌发生的潜在分子机制仍未完全阐明。

糖酵解是由一系列酶催化的级联反应过程,催化糖酵解的第一个关键酶就是己糖激酶(Hexokinases, HKs)。HK催化进入细胞内的葡萄糖磷酸化生成6-磷酸葡萄糖(G-6-P),并消耗一分子ATP。现已发现的人类HK有4种亚型,分别由HK1、HK2、HK3和HK4基因编码生成。HK1广泛表达于几乎所有的哺乳动物组织中,HK2通常表达于胰岛素敏感组织如脂肪、骨骼和心肌中。HK3则往往低水平表达,HK4的表达则限制于胰腺和肝脏。

近年来的研究发现在一些癌细胞中HK2呈高水平表达并且与癌症患者不良的总生存率相关。一些小鼠模型遗传研究证实,HK2在Kras驱动的肺癌和ErbB2驱动的乳腺癌中发挥关键作用。

在这篇文章中,通过对Pten-/p53缺失小鼠胚胎成纤维细胞(MEFs)、前列腺癌细胞系、异种移植物、遗传工程小鼠模型以及临床前列腺癌样本展开分析,研究人员证实在前列腺癌细胞中Pten和p53缺失选择性上调了HK2。进一步的机制研究表明,Pten删除激活了AKT-mTORC1-4EBP1信号轴,而p53缺失通过抑制miR143生物合成提高HK2 mRNA稳定性,由此增进了HK2 mRNA翻译为蛋白质。一些遗传研究证实,HK2介导的有氧糖酵解(称作为Warburg效应)是在前列腺癌异种移植物小鼠模型中Pten-/p53缺失驱动肿瘤生长的必要条件。

这些结果表明了,HK2有可能是携带Pten和p53突变的前列腺癌患者的一个潜在治疗靶点。

1997年毕业于白求恩大学获医学博士,1998-2002年间先后在纽约大学和美国休斯顿贝勒医学院从事博士后工作,之后在休斯顿德州大学安德森癌症中心遗传学部门担任助理教授,现为美国明尼苏达大学癌症遗传学实验室主任。在人类癌症细胞死亡通路、P53对线粒体的调节和TNF对细胞死亡受体的调节等细胞死亡通路方面研究具有巨大成就。近年来,先后以第一作者或者通讯作者在Cell、Nature、PNAS、EMBO J、MCB、Genes &Dev等顶尖一流杂志上发表论文10多篇。
