Toxicol Sci:mmu_piR_003399在调节微囊藻毒素MC-LR诱导的生殖毒性中起关键作用

研究提示微囊藻毒素MC-LR对雄性小鼠具有毒性作用,对精原细胞具有细胞毒作用,并导致小鼠睾丸的组织和功能发生改变。

据报道,已有五、六千种化学物质具有生殖毒性。二溴氯丙烷是典型的生殖毒物,可致精子减少,活力缺乏和性腺发育不全,以致不育。镉、邻苯二甲酸二乙基已酯可引起不同类型的睾丸损害,多环芳烃、博来霉素和二硫化碳可致雌性生殖系统的损害。研究提示微囊藻毒素MC-LR对雄性小鼠具有毒性作用,对精原细胞具有细胞毒作用,并导致小鼠睾丸的组织和功能发生改变。因此研究人员调查了P元件诱导的Wimpy(piwi)相互作用RNA(piRNA)在MC-LR诱导的雄性小鼠生殖毒性中的功能。

研究显示,用MC-LR处理后精原细胞和小鼠睾丸组织中mmu_piR_003399水平增加。细胞周期蛋白依赖性激酶6(CDK6)是mmu_piR_003399的靶基因,mmu_piR_003399浓度的增加与体外和体内CDK6的表达降低相关,mmu_piR_003399诱导G1期细胞周期停滞,下调精子数量和精子活力,并影响精子形态。相反,抑制mmu_piR_003399的表达可显著减弱MC-LR诱导的小鼠病理学改变,包括细胞周期停滞,成熟精子数量减少,精子活力损失和精子形态异常。研究数据显示,mmu_piR_003399存在于小鼠血清和血浆中,并且其水平在MC-LR处理的小鼠中增加。

该研究结果表明,mmu_piR_003399在调节MC-LR诱导的生殖毒性中起关键作用。
