最近,来自德国环境健康研究中心的研究人员研究发现内燃机产生的纳米颗粒可以激活肺部组织细胞中休眠的病毒,相关研究成果近期发表在《Particle and Fibre Toxicology》杂志上。

为了躲避免疫系统,许多病毒都隐藏到了宿主细胞中且长期存在。如果免疫系统变弱或者产生某些条件,这些病毒就会再次活化、增殖并损伤宿主细胞。来自德国环境健康研究中心的Tobias Stöger和Heiko Adler博士领导的一个科学研究团队发现纳米颗粒也可能促使这个过程发生。

“过去的研究已经表明吸入纳米颗粒会导致炎症并改变免疫系统。”研究领导者Stöger博士说道。他和他的同事Heiko Adler及Philippe Schmitt-Kopplin教授一起研究发现肺暴露在纳米颗粒中可以再次激活其中的疱疹病毒。

他们在一个特定的疱疹病毒感染模型中检测了化石燃料内燃机产生的纳米颗粒的影响。他们发现病毒蛋白明显增加,而这些蛋白只能由活化的病毒产生。“代谢和基因表达分析也发现了和急性感染相似的现象。” Philippe Schmitt-Kopplin教授说道。此外,他们在人细胞进行进一步实验发现这些细胞接触这些纳米颗粒后,潜伏其中的埃博斯坦病毒也被唤醒。

在未来的研究中,研究团队将检测这些现象是否也在人体内发生。“许多人也携带疱疹病毒,患特发性肺纤维化的病人受这些病毒的影响更严重。”Heiko Adler说道,“如果在人体内也观察到相同的现象,那么研究吸入的微粒激活潜伏疱疹病毒的分子机制就显得至关重要。我们将会尝试用治疗性方法去干预这些分子信号通路。”

特殊的细胞培养模型可能帮助阐明纳米颗粒激活病毒的精确机理。“此外,我们将在长期研究中调查纳米颗粒的反复暴露及由此激活的病毒将如何导致慢性炎症及肺部重塑。”
