Nat Commun:新研究简化小鼠换血方法

11月23日,《自然—通讯》期刊介绍了一种年轻小鼠与年老小鼠换血新方法。研究显示,在不同年龄段的小鼠之间换血,可以在短短几天之内改变小鼠组织功能。这种输血系统可能有助于研究组织老化。

过去研究显示,年轻血液成分可以使衰老组织重获新生,而老化血液会对年轻组织产生有害影响。但这些实验通常依赖于异时骈体方法,通过这种复杂的手术,两种动物的循环系统被连接在一起,并维持几个星期的时间。

它们除了共享血液,还会共享其他身体组件,包括器官,这意味着年老小鼠可以获得年轻小鼠的肺、免疫系统、心脏、肝和肾,而年轻小鼠必须额外维持一具衰老的身体。美国加州大学伯克利分校的Irina Conboy及同事设计了一种简化的输血系统,仅在小鼠之间交换血液,借此消除共享器官对换血效果的影响。

该研究团队表明,换血后6天就显现出了效果:年轻血液可以增强受损老化肌肉的修复,而老化血液会抑制年轻小鼠体内新脑细胞的形成和肝细胞再生。整体而言,老化血液的抑制作用比年轻血液的有益作用更明显。

作者认为该研究所述换血系统比异时骈体多几项实践优势,例如速度更快、侵入性更小,而且能够推动未来在有力的控制情况下,研究识别影响组织老化或再生的血源因素。
