研究认为Lenti-D慢病毒载体基因治疗可有效缓解早期X-连锁肾上腺脑白质营养不良患者的病情，是异基因造血干细胞移植之外的一个治疗选择

X-连锁肾上腺脑白质营养不良患者中，ABCD1基因突变导致ALD蛋白功能丧失。肾上腺脑白质营养不良的特点是脱髓鞘和神经退行性疾病，导致神经功能丧失和死亡。目前异基因造血干细胞移植是临床中普遍的治疗方法。

在这项II-III期临床研究中，研究人员招募肾上腺脑白质营养不良男童，患者处于疾病早期且通过钆增强磁共振成像（MRI）筛查，患者接受Lenti-D慢病毒载体自体CD34细胞转导治疗。中期分析中评估移植物抗宿主病、死亡、主要功能性残疾的发生率，以及神经功能的改变和MRI病变程度。研究的主要终点为存活率以及治疗后24个月的功能障碍。

17名男童接受Lenti-D基因治疗，随访29.4个月的中期分析显示，所有患者植入后基因标记细胞阳性，无已知癌基因或克隆生长产物优先整合的证据，所有患者ALD蛋白检测成阳性，无治疗相关死亡或移植物抗宿主病报告。17名患者中，15人(88%)存活，无主要功能障碍且临床症状轻微。1名神经功能快速恶化病人死于疾病进展，1名MRI疾病进展病人退出研究，接受异基因造血干细胞移植后死于移植相关并发症。
