蛋白质甲基化是一种非常重要的翻译后修饰现象，其甲基供体为S-腺苷甲硫氨酸（SAM）。由于甲基化可发生在氨基酸残基的N-,O-和S-中心，并且N-中心甲基化还存在不同状态，同时甲基化对分离行为影响非常细微，限制了对蛋白质甲基化组的解析。常规基于抗体的研究策略仅捕获了部分N-中心甲基化的信息。中国科学院大连化学物理研究所研究员赵宗保和邹汉法提出了“同位素标记SAM甲基化”研究新策略，深入解析了模式生物酿酒酵母的蛋白质甲基化组，首次对蛋白质甲基化实现全覆盖，研究成果最近发表在Journal of Proteomics(2015, 114, 226)上。

该成果成功鉴定了43种甲基化蛋白，包括分布在64个甲基化中心的68个甲基化事件，其中超过90%的甲基化事件是首次发现。分析表明：（1）超过2.6%的酵母蛋白质存在甲基化；（2）甲基化发生在氨基酸残基的顺序为Lys >> Arg > Asp > Glu ≈ Asp ≈ Gln ≈ His > Cys；（3）N-甲基化状态存在互斥性。上述结果极大地丰富了对蛋白质甲基化的科学认识，为研究甲基化蛋白质的功能提供了重要信息。
