血管内皮蛋白酪氨酸磷酸酶抑制可使肿瘤血管的结构和功能正常化。 实体瘤结构和功能异常的特征在于其微血管形成,并且微血管形成介导了数种有害的肿瘤行为。血管内皮蛋白酪氨酸磷酸酶(VE-PTP)是一种抑制内皮细胞Tie-2受体酪氨酸激酶的物质,在本研究中,为了确定VE-PTP对阻碍肿瘤血管成熟所起的作用,来自美国哈佛医学院的Shom Goel等设计了相关研究,研究结果发表在JNCI 8月的在线期刊上。

研究要点:

1.实体瘤中的微血管存在功能缺陷且不成熟,这会加速肿瘤的转移以及降低抗肿瘤治疗的效果。

3.AKB-9778是一种新型的、强效的、选择性的VE-PTP抑制剂,可起到稳定肿瘤血管和减轻肿瘤血管不稳定所带来的不利影响的作用。

AKB-9778是VE-PTP抑制剂。研究者通过离体实验和使用斑马鱼胚胎血管形成的载体实验确定了VE-PTP对ECs的效果。研究者应用小鼠乳腺癌模型及自发转移和试验转移模型就AKB-9778对肿瘤血管形成、肿瘤生长和转移进展的效果进行了研究。最后研究者采用内皮一氧化氮合酶(eNOS)缺乏的小鼠来确定eNOS在介导VE-PTP抑制方面所起到的作用。所有的统计检验都在双侧进行。

AKB-9778诱导ECs中的配体非依赖性Tie-2的激活,会阻碍斑马鱼胚胎的血管生成。AKB-9778通过维持血管的成熟度来延迟早期乳腺肿瘤的生长;通过预防肿瘤细胞外渗来减慢微转移的速度,从而可以延长患者的生存期。AKB-9778还能稳定原发肿瘤血管、增加肿瘤灌注和对放疗的反应。内皮一氧化氮合酶部分介导AKB-9778对肿瘤血管形成的作用。

本研究结果证实了VE-PTP抑制剂能使肿瘤血管的结构和功能正常化,这主要是通过激活Tie-2实现的,从而可以延缓肿瘤的生长、减慢肿瘤转移进展,以及增强对同时所进行的细胞毒治疗的应答。

研究背景:

与健康组织中的血管不同,实体瘤中的微血管是存在功能缺陷和不成熟的。肿瘤的血管存在结构和功能的不稳定,这会促进肿瘤进展过程中的数个独特的阶段的出现,包括早期的血管芽生、肿瘤细胞转移到远处的器官、以及肿瘤缺氧,这些都会加速肿瘤的转移以及降低抗肿瘤治疗的效果。

内皮细胞受体酪氨酸激酶Tie-2/Tek是一种调节血管成熟的关键调节剂。Tie-2经由其血管生成素1配体拮抗剂(Ang-1)所激活,从而增加血管周围细胞的覆盖、加强EC结合、降低通透性、以及增加血管的直径。最近的研究显示Ang-2在介导肿瘤血管异常和肿瘤转移过程中所起到的作用,这提示Ang-2所造成的Tie-2失活部分导致了肿瘤内不稳定血管的出现。

目前靶向针对Ang/Tie-2轴的治疗策略主要是直接针对血管生成素的靶向治疗,但是这类研究并没有直接评估Ang-1和Ang-2之间的相互作用,也没有评估血管生成素阻滞对Tie-2活性的影响。目前在临床研究中约有10种靶向针对Ang/Tie-2轴的制剂。

血管内皮蛋白酪氨酸磷酸酶(VE-PTP)是一种EC特异性受体酪氨酸磷酸酶,其具有去磷酸化的作用,能够使Tie-2失活。但是VE-PTP在肿瘤血管生成中所起到的作用还属未知。最近,研究者们研发了AKB-9778,其是一种新型的、强效的、选择性的VE-PTP抑制剂,主要通过调节1,2,3,4-四氢异喹啉氨基酸的结构来实现上述作用。我们假设VE-PTP的抑制能不通过Ang-1/Ang-2配体的通路来激活EC中的Tie-2信号通路,从而稳定肿瘤血管和减轻肿瘤血管不稳定所带来的不利影响。
