I型干扰素的产生I型干扰素是参与抗病毒免疫的重要效应分子。它的产生主要由先天性免疫细胞(主要是巨噬细胞)表面或内部受体(Toll like receptor,NOD like receptor,RIG-I receptor,cGAS等)接触到病毒特异性的抗原物质(DNA,RNA),然后通过胞内的信号分子传递(MAVS,STING,TBK,IKK等),最终激活转录因子IRF3/7从而启动I型干扰素基因的表达。
 
I型干扰素的信号传递:I型干扰素是分泌型蛋白,它们的受体是一类位于细胞膜上的异源二聚体(由IFNR1与IFNR2两个亚基组成)。配体受体的结合后会激活下游的蛋白激酶JAK1与TYK2,激酶活化后会激活胞浆内的转录因子STAT1与STAT2,俩者聚合进入细胞核内协助IRF9转录下游的一些效应基因。
 
I型干扰素的抗病毒免疫保护效应:1、体外研究普遍证明,IFN通过促进下游一些ISG(IFN stimulated gene)的表达,从而达到一致病毒复制的能力。这些基因包括MX1,PKR,OAS,IFITM,APOBEC1,TRIM等。2、同样,体内试验也证明:当小鼠缺失IFNR1/2时,对疱疹病毒,生里基森林病毒,豆苗病毒,脑膜炎病毒等感染失去抵抗能力。以最受关注的免疫缺陷病毒为例:通过对恒河猴进行SIV感染(相当于HIV),发现在阻断IFN信号通路后,病毒库容积增大,CD4+T细胞加速减少,加速了艾滋病的感染效率。如果此时向恒河猴体内注射重组I型干扰素则会提高抗病毒基因的表达并抑制病毒的系统性感染。然而,持续的给药则会使宿主"脱敏",抗病毒基因的表达量随之下降,艾滋病随即继续加速恶化。说明I型干扰素的表达时间与剂量是十分关键的。3、一些研究发现I型干扰素能够促进树突状细胞的活化,提高细胞表面MHC-II与辅助刺激因子CD80,CD86的表达,从而增强其抗原呈递的能力。另外,一些研究认为I型干扰素可以直接作用于CD4+与CD8+T细胞以及B细胞,提高它们的活化水平。
 
I型干扰素的负面效应:然而,最近的一些研究发现I型干扰素可能会导致病毒感染中的免疫抑制以及炎症反应,从而使疾病发生恶化。在SIV感染猿猴的模型中,研究发现强烈的I型干扰素反应在病情恶化的动物体内比较常见,而病情稳定的动物体内I型干扰素的表达量相对较低。进一步研究发现,I型干扰素会引起细胞表达TRAIL(一类细胞凋亡受体),从而引起细胞的死亡。免疫细胞的死亡则会促进病毒的复制与感染。另外,在急性感染中,I型干扰素的过量表达会引起细胞的凋亡与组织的损伤,触发炎症反应,从而使疾病发生恶化。
 
此外,I型干扰素在抗细菌及抗真菌感染中也有一些正面与负面的效应,因为不太重要,所以不再赘述。
 
综上,我们了解了I型干扰素的产生机制与作用机制,以及其在抗病毒免疫过程中的主要作用于负面影响。希望这些信息能够帮助我们开发抗病毒感染的I型干扰素药物。
