PNAS:红细胞成熟机制新发现——有望治疗特定类型贫血

在红血细胞发育过程中,骨髓干细胞需要经过几个阶段直到最终激活血红蛋白和其他红细胞的相关蛋白的基因,从而得到成熟红细胞。为了在实验室中模拟这一过程,研究人员先前发现,在培养血清中加入血细胞祖细胞有助于激活所有相关蛋白质,从而最终得到红细胞。

现在在西湖高级研究学院和北京大学的Gao和Lee想知道血清中是否存在重要的物质可以作为开关使细胞转换为一个成熟的红细胞。

为了探寻到血清中的哪种分子是始发因素,Gao和Lee通过一种标准实验室过滤器来运行血清,这种滤器在现实生活中很容易见到,主要用来过滤自来水,就是木炭。

木炭以吸附空气中的气味和水的味道而闻名。研究人员注意到,一旦过滤,血清不再支持红血细胞的生产;他们推断其中的疏水性分子甲状腺激素被活性炭吸附走, Gao和Lee认为只要将甲状腺激素加回到血清中、红细胞祖细胞就会再次启动红细胞的成熟过程。

甲状腺激素在刺激红细胞成熟过程中起着非常重要的作用,他们发现,如果在早期发育阶段加入甲状腺激素,红细胞会缩短其正常发育过程,并开始变成成熟的红细胞。

Gao和Lee随后探索了甲状腺激素影响红细胞成熟背后的具体机制。最终他们找到了特定类型的成熟红细胞的胞内受体,可以和甲状腺激素结合。

通过从这里发现的甲状腺激素和红细胞成熟之间的关系,科学家可以探索新的刺激红细胞生成的治疗方法来治疗特定类型的贫血患者。
