科学家对细菌基因组成功实现了根本性地“重写”

近日,在Science杂志上刊登的一篇文章中,合成生物学家报道了一种对细菌基因组进行重新“布局”(rewiring)的突破性研究成果,这项研究报告中,研究人员对大肠杆菌基因组中3.8%的碱基对进行了“再利用”,并且成功改变了其用途。

科学家将64个遗传密码子中的7个替换成了能够产生相同组分的遗传密码子,遗传密码子是可以编码产生氨基酸的基因序列,同时研究者通过在55个DNA片段中合成DNA就减少了密码子的数量,而每一个片段都是长为5万个碱基对的DNA片段,随后研究人员利用这些片段重新组装形成了功能性的大肠杆菌(E.coli)。

尽管如此,来自哈佛医学院的研究者表示,这是我们向开发具有新型特性的工程化有机体迈出的巨大一步,比如我们开发抵御病毒感染的有机体等。包括George Church在内的多个合成生物学家表示,这项工作为人类基因组计划的进展提供了一种研究原型,在人类基因组计划研究中,科学家们的终极目的就是成功合成人类的基因组。

大的改变

研究者Church说道,这项研究表明,这种根本性的重组技术或许是可行的;来自耶鲁大学的研究者Farren Isaacs认为,在自然界中将有机体密码子从64个减少为57个是一项非常大的壮举,如今我们取得了巨大成功,而这主要表现在我们阐明了遗传密码子的可塑性,同时还揭示了如何通过对基因组的重新编码来从有机体中提取出全新类型的生物学功能和特性。

研究者Church的实验室和其他研究者此前通过研究发现,他们有可能对大肠杆菌的单个氨基酸进行重新编码,以便细菌可以插入在自然中找不到的氨基酸分子;而诸如这样的重编程有机体或许会对病毒感染产生较强的耐受力,因为重组的有机体中不再含有所有自然界生物体中所常见的遗传机器,而病毒往往会利用有机体中的常见遗传机器来进行自我复制。这些重编程有机体的制造主要依赖于饮食中合成性的氨基酸,同时其还能缓解科学家们的担忧,而科学家们会担心重编程的细菌会“逃离”实验室并在自然界开始肆虐。

我们所拥有的先进技术

研究者在最新的研究中使用的重编码过程是一项非常艰难的开发过程,多年前研究者根本不可能看到这种技术,然而在过去10年里,工程学研究和合成性DNA技术的速度大大加快,从而就促进科学家们开展了很多遗传工程学新技术及研究。

来自华盛顿大学的研究者Marc Lajoie说道,这种遗传工程学计划目前正处于前所未有的大规模研究之中,而且如今我们能够开发出大型完整的合成性基因组,同时我们还能够将最多的功能性改变引入到合成性的基因组中去。

来自雷格文特尔研究所的研究者Craig Venter今年3月份宣布,他们以所有不必要的基因被移除的细菌基因组为基础开发出了一种合成性的基因组,但有机体的基因组在数量级上往往要小于大肠杆菌。如今研究者Church和其同事正在尝试将重编码大肠杆菌的DNA片段“缝合”到连续的基因组上,随后他们将检测是否这种重组的有机体是否具有生存的能力;Church表示,我们并不清楚这项计划需要多长时间才能完成,实验室的其他同事则推测这项研究需要持续数月甚至数年。
