STM:仿生耳技术可使听力神经再生

携带有耳蜗移植物的病人能听讲话,但对音调的感知却很糟糕,更不能享受音乐。研究人员希望这些依赖耳蜗移植物的患者能感受更广范围的动态的声音。长久以来一直认为当神经营养因子(Neurotrophins)运输到听力内耳即耳蜗后,听力神经在这种对神经元发育、功能和生存非常重要的蛋白质家族的作用下会丧失再生功能。而且,用药物安全地靶向运输神经营养因子几乎不可能,基于病毒的基因治疗也无能为力。

该研究团队用现有的耳蜗技术进行基因治疗,刺激因年龄以及环境因素导致失聪后存活的听力神经的再生功能。这种利用耳蜗移植物的电流脉冲运输DNA到移植物电极周围细胞的方法能使这些细胞重新生产营养因子。几个月后当营养因子的产量下降后,由此产生的神经活性将最终改变听力神经。这种移植物在动物模实验中获得成功。研究人员推测如果能够诱导移植物电极周围听力神经的再生并加强它们之间的联系,那么这种方法将在人体成功。

研究人员指出,将来这种基因靶向治疗所需时间短,在移植手术中外科医生安装好移植物后只需将DNA注入耳蜗并启动电流脉冲即可。这种技术的应用不局限于听力障碍的治疗,当这种技术与其他仿生设备结合时,如结合治疗帕金森症和抑郁症的大脑内部电极阵列,则可直接进行安全地靶向基因治疗。
