近日，来自伊利诺伊大学的研究人员在Nature Materials杂志上发表论文指出，恶性肿瘤中仅有少数细胞会逃脱并且扩散到机体其它部位，而肿瘤边缘的曲线在激活这些肿瘤种子细胞（转移性肿瘤细胞）上扮演着重要作用。文章中研究者在多种形状和模式中利用工程化的组织环境对皮肤癌进行研究，他们发现，细胞的培养物越弯曲，其边缘的癌细胞就越会表现出干细胞的特性，而这种特性是扩散到机体其它位点的关键。

研究者Kristopher Kilian表示，癌症最危险的部分就是发生转移，我们把有些细胞称之为癌症干细胞，其会采用致命的特性，顺着血管不断“游动”从而在新位点处形成新的肿瘤，目前我们需要一种新方法来发现这些癌症干细胞并对其进行研究，当然更重要的是开发新型药物来对其进行靶向作用，目前癌症干细胞对化疗药物已经产生了耐药性，而这是引发患者癌症复发的主要原因。

文章中，研究人员在多种不同形状和模式的2-D和3-D环境中对皮肤癌细胞进行了培养，来观察是否肿瘤的形状会帮助激活癌症干细胞，以及癌症干细胞在肿瘤中出现的位置；结果发现，癌症干细胞会沿着工程化肿瘤环境的边缘大量产生，尤其是在肿瘤环境的转角和凹凸面。正常的干细胞喜欢柔软、湿润的组织内部位置，而癌症干细胞则不然，随后研究人员对多种癌症细胞系进行了类似研究，结果发现，不同的癌症干细胞均会以相同的方式来对模式化的肿瘤环境产生反应。

研究者Kilian希望这种模式工程化的组织环境会给科学家们提供一种方法来发现并且培养出癌症干细胞，他说道，我们可以想象一位患者机体有着特殊的肿瘤，而我们可以在培养皿中对其进行工程化操作，并且利用患者自身的细胞来开发出特殊的肿瘤模型进行特殊靶向药物的检测，这或许可以帮助我们尽快找到靶向治疗特殊癌症患者的新型疗法。

研究者表示，后期还有大量工作要开展，但本文的研究成果对于后期新型药物的开发、手术的进行以及揭示癌症的进展及扩散机制都提供了一定的研究基础；癌症是非常复杂的，因此研究癌症发生的方方面面非常关键，如果有一种微环境可以帮助激活促进癌症转移的癌细胞，那么我们或许就可以通过阻断这种微环境来间接阻断癌症的转移和进展。
