肥胖可通过影响卵巢环境扰乱卵母细胞成熟进而影响胚胎发育

近日,内分泌领域权威杂志JCEM上发表了一篇临床研究,研究人员旨在比较接受生育治疗的超重/肥胖女性和体重正常女性的卵母细胞的基因表达和卵泡液含量。

来自美国密歇根州立大学的Meghan L. Ruebel及其同事招募了接受生育治疗的11名超重/肥胖女性和 13名体重正常女性。他们通过单细胞转录组测序分析卵母细胞的基因表达情况,并采用酶联免疫吸附试验(Elisa)和比色法对血清和卵泡液进行了定量分析。

本研究发现:和体重正常的女性相比,超重/肥胖女性表现出较高的体重指数(BMI)、体脂百分比和血清HOMA指数(p<0.01),且瘦素、C反应蛋白、卵泡液瘦素以及甘油三酯含量均显著增加(p<0.05)。此外,超重/肥胖女性的卵母细胞中的促炎因子(CXCL2, p=0.071)及氧化应激相关基因(DUSP1, p=0.051)的表达增加。然而,GAS7 (脂质代谢相关基因, p=0.065)、TXNIP (氧化应激相关基因, p=0.055) 以及转录因子ID3 (p=0.075) 和TWIST1(p=0.09) 的表达是下降的。

该研究说明:对于接受激素诱导卵母细胞治疗的女性而言,身体组成可以显著影响卵母细胞的转录丰度。该研究进一步确定了产妇饮食(特别是饱和脂肪酸的摄入)对卵母细胞基因表达具有潜在的影响。因此,孕前是进行生活方式干预的一个关键时期。
