SCAPs培养于成牙本质分化的条件,并额外施加ASA与否。也将施加ASA刺激的SCAPs植入裸鼠皮下。

结果显示,ASA体内外加速SCAPs的成牙本质分化,并下调Runx2,上调SP7、核因子I C及DPP表达。ASA上调成牙本质分化的SCAPs内AKT磷酸化水平。有趣的是,施加磷酸肌醇酯3-激酶抑制剂LY294402和siRNA AKT促进体内外ASA诱导的SCAPs成牙本质分化。LY294402和siRNA AKT也抑制SCAPs 内ASA诱导的Runx2表达,并增强ASA诱导的SP7、核因子I C及DPP表达。

结论:这些结果表明,施加ASA及抑制磷酸肌醇酯3-激酶-AKT信号通路的调控是基于SCAPs牙根再生的一种全新方法。
