根尖牙乳头来源的干细胞(SCAPs)拥有较强的成牙本质/骨分化能力。这篇研究的目的是为了评估环磷腺苷(cAMP)对SCAP成牙本质/成骨分化的影响以及cAMP与TGF-β1之间潜在的相互作用。研究对施加了TGF-β1或TGF-β1抑制剂的SCAPs施以cAMP激活剂（福司柯林）刺激。检测钙矿化沉淀量以及碱性磷酸酶活性的变化。实时定量聚合酶链式反应评估cAMP对TGF-β1介导的SCAP成牙本质/骨分化情况。采用WB分析cAMP对TGF-β1诱导的Smad2/Smad3和ERK/P38磷酸化水平的影响。结果显示，施以TGF-β1抑制剂和福司柯林能够增强SCAPs的钙矿物质沉积和碱性磷酸酶活性。TGF-β1抑制剂协同福司柯林对I型胶原和Runx2表达的影响。WB结果显示，福司柯林减弱TGF-β1诱导的Smad3和ERK磷酸化水平，而cAMP抑制剂(H89)可以拮抗这种作用。结论：这篇研究表明，cAMP信号通过抑制Smad3和ERK磷酸化水平干扰TGF-β1信号，进而促进SCAPs的成牙本质/骨分化。原始出处：Zhang J， Zhang CF， et al.
