近日刊登在Nature上的一项研究报告中,来自斯坦福大学医学院的研究者通过研究鉴别出了一种特殊的细胞标志物,其可以帮助研究者更好地挑选出可以产生血液及免疫系统的最基本形式的干细胞,一旦本研究被证实,那么这或许可以帮助解决长期以来关于鉴别基础干细胞及支持细胞的争议了,同时也将为理解基础干细胞自我更新的机制提供新的思路。

文章中,研究人员认为他们找到了一种可靠地方法来区分长期和短期的造血干细胞的差异,这种在骨髓中存在的特殊单一标志物目前仅在长期的造血干细胞中被发现,基于此研究者就可以有效鉴别出长期的造血干细胞了,他们希望可以揭示这些细胞及附近的细胞如何制造一种维持干细胞的特殊生境。研究者James Chen说道,近乎30年来科学家们一直在寻找如何在体外培养生长造血干细胞,但都一直没有实现,如今我们开发了特殊的方法来解决这个问题。

过去10年里很多科学家们都提出了针对长期造血干细胞而言的多种特殊的标志物,但针对每一种标志物的可靠性不同科学家们之间产生了激烈的争论;为了解决这个问题,研究者Chen及Miyanishi设计了一种高度系统性的方法,随后他们对在骨髓中(造血干细胞也存在于骨髓中)进行表达的100多个基因进行了研究,研究者们剔除掉了骨髓中不能产生血液和免疫细胞的基因,最后将范围限定到了45个基因;他们进行了一项复杂的分析揭示了这些基因在多种细胞中可以制造出的蛋白质的量,结果发现,仅有三种蛋白可以达到足够高的水平来制造造血干细胞,进一步筛查研究者们最终发现仅有一种基因符合要求,即为Hoxb5。

研究者认为,在长期的造血干细胞中或许还存在其它特殊的标志物,比如在最早筛查的100个基因外的某个特殊基因,但在本文中所筛选的基因中仅有Hoxb5是长期造血干细胞存在的特殊基因标志物。鉴别长期干细胞的能力可以为科学家们提供一种强有力的工具,本文研究也可以帮助科学家们在生境中有效获得长期造血干细胞和其它的细胞。
