抗老+抑癌,尚未产业应用的诺奖新秀会被催生出泡沫吗?

(1945年2月9日-),日本分子细胞生物学家。现任综合研究大学院大学名誉教授、基础生物学研究所名誉教授、东京工业大学前沿研究机构特聘教授。 大隅教授是细胞自噬研究的先驱,曾获得京都奖、盖尔德纳国际奖及日本人第2座威利奖。2016年获得日本人第4座诺贝尔生理学或医学奖。

几个小时前,2016年诺贝尔生理学或医学奖揭开面纱,日本分子细胞生物学家、京都工业大学大隅良典因其在自噬性溶酶体方面做出的贡献而摘得桂冠,并获得奖金800万瑞典克朗,约合625万人民币。

大隅良典的研究侧重于酵母细胞自噬,在该研究中他“朝着阐明自噬的分子机制和生理意义,作出了开创性的贡献。”(该点评来自于2012年京都奖,大隅良典是那一年生命科学相关的京都奖唯一获奖人,在此之前,2010年诺奖得主山中伸弥在2010年也曾获得了京都奖)。

在诺奖公布的获奖原因中,对于大隅良典的研究贡献官方是如此表述的:他的工作不但揭示了一种基本的细胞基质,更多许多疾病的阐明铺平了道路。

不过相比较去年屠哟哟获奖的“满屏涨停”此次诺贝尔医学奖恐怕会使二级市场投机者失望了,相比较此前被媒体和学界高度猜测的基因编辑以及肿瘤免疫治疗,细胞自噬目前在全球范围尚未触及真正的临床产业应用,仍处于研究阶段,并且距离产业技术的落地还有比较遥远的一段距离。

“细胞自噬比较类似细胞凋亡,凋亡和自噬是细胞死亡的两种途径。它是一个科学研究的现象,结合大隅良典研究的这个途径,有可能在一些疾病的发生发展过程中起作用,从而产生药物开发的靶点。”上海东富龙医疗科技产业发展有限公司总经理劳昕元表示。

“但是细胞自噬本身并不是一个平台级的技术,所以不会有很大的产业热点,因为它无法在某个领域马上应用去治病。相对来说,免疫治疗和基因编辑虽然还没有获得诺奖,但这两个技术在产业届却是多面手。有很多实际临床应用的案例,所以在热点度上来说,细胞自噬肯定是差很多的。”

另一位医疗领域投资人高特佳投资合伙人王海蛟也对表达了相似的观点,“在目前国内上市公司的布局中,还没有发现与之相关的产业化研究。细胞自噬在抗衰老和帕金森相关疾病的的应用机制上研究更早一些,而抑制肿瘤领域目前是一个潜在的研究方向。”
它会如何对抗衰老?

大隅良典曾研究表达自噬蛋白的基因


“和细胞自噬产业化相关的最主要是关注与药物相关的靶点和临床进度,它是一个现象,目前并没有直接很广泛的应用。”劳昕元说。

自噬,是一种细胞处于压力之时消化蛋白质和细胞器的自适应过程,以下几种情况会导致自噬现象的发生:
第一种情况是异源自噬,当一些感染性的例子进入细胞后,细胞就会启动自噬程序,清除异物,这在细胞消灭病原体上有着重要意义;

第二种情况是细胞的自我修复,在运作一定时间后,一些细胞就会出现受损。自噬能够吃掉这些旧的细胞器,并把消化后产生的大分子原料用于新细胞的合成。

细胞自噬近年来受到热捧的一个很重要原因就在于这些机理与疾病的关联,其中,首当其冲便是对抗衰老


细胞自噬的典型特征是形成自噬体并呈递给溶酶体,这一过程在蛋白质和细胞器质量控制中起基础作用并维持了细胞能量的稳态。一些研究表明,自噬与细胞衰老密切相关,参与蛋白酶和自噬相关调节的BAG蛋白家族中BAG3/BAG1比值在复制性衰老时增高,且BAG3在细胞衰老时能介导自噬的激活。研究还发现在Ras诱导的细胞衰老进程中亦可观察到较高的自噬活性。

除此以外,对于抑制肿瘤细胞自噬也让学界充满期待,近年来大量的研究表明,自噬与肿瘤的发生发展密切相关,许多调节自噬的药物被用于临床肿瘤治疗的研究中。

不过,对于许多细胞生物学的科研人员来说,相比较细胞自噬,细胞凋亡这个名词可能更为亲切:2002年“细胞凋亡”就已经获得了诺贝尔生理/医学奖,而这次,14年后,与之一脉相承的细胞自噬再次获得了诺奖评审委员会的属意。

根据科研公开资料,“细胞凋亡”(又称“程序性细胞死亡”)规律的发现曾让人们更好的了解了生老病死的客观规律,而如今,细胞自噬恐怕将会赋予临床更多的探索意义:它可以帮助人类更有效的延缓衰老、攻克肿瘤疾病,这些临床应用的领域包括癌症、帕金森症、阿尔茨海默以及心肌病。
