Novavax新冠疫苗寄予厚望,后起之秀疟疾疫苗迎来里程碑

然而, Novavax在南非进行的另一项2b期临床试验中期结果显示,该疫苗的确有效,但效果却不及针对英国的这种疫苗。南非的研究包括一些艾滋病毒志愿者。在艾滋病毒阴性的志愿者中,这种疫苗似乎有效率为60%。若包括艾滋病毒志愿者在内,总体上该疫苗有效率仅为49.4%。到目前为止,在南非研究中发现的90%的新冠病例是由于新变异毒株引起的。

4月23日,牛津大学Mehreen研究团队在《柳叶刀》杂志在预印本上在线发表了评估疟疾候选疫苗R21的2b期临床试验的结果。结果显示该疫苗的有效率为77%。

该研究招募了来自名为Nanoro的地区的450名参与者,季节性疟疾传播率很高。在三个研究小组中,年龄在5至17个月的参与者接受了5mg R21、25mg或50mg Matrix-M佐剂或狂犬病疫苗(对照)。参与者每四周间隔接受三剂,一年后接受最后一剂第四剂。对该疫苗的安全性,免疫原性和功效进行了一年以上的评估。

研究人员在文章写道,在较高的辅助剂量组中,六个月的疫苗效力为77%,在较低的辅助剂量组中为71%。一年后,高辅助剂量组的疗效保持在77%。这大大高于迄今为止最有效的疟疾疫苗候选者RTS,S / AS01疫苗,在非洲儿童中,该疫苗在12个月内的有效率为55.8%。

从2b阶段的结果来看,Matrix-M似乎可以帮助提高功效非常明显。在这项研究中,给17个月至5岁的儿童服用5毫克R21和25毫克或50毫克的Matrix-M。较低的Matrix-M剂量可达到71%的功效,而较高的剂量则可达到77%的功效。

据报道,两种佐剂的剂量水平都耐受良好,没有严重的反应。此外,接种R21 / Matrix-M的参与者在第三次接种后28天显示出高滴度的疟疾特异性抗NANP抗体,在较高的辅助剂量下几乎翻了一番。尽管抗体滴度会随着时间的流逝而减弱,但是在一年后的第四次给药后,抗体的滴度提高到了与初次接种一系列疫苗后达到的峰值滴度相似的水平。
