Stroke:胱抑素C是卒中的内源性神经保护因子!

胱抑素C是导致内源性神经保护的一个重要因素。这也是一种新的通过维持溶酶体膜完整性的卒中候选治疗手段。

内源性神经保护可由缺血和非缺血预处理来诱导。然而,并不是所有接受这些预处理的受试者可以得到类似的有利结果。近日,卒中领域权威杂志Stroke上发表了一篇研究文章,这项研究旨在探索这一现象的分子机制,并试图找到新的卒中治疗靶标。

研究者将成年健康的雄性SD大鼠进行短暂性大脑中动脉闭塞手术,通过高通量蛋白质组学技术,以及通过等压标记进行相对和绝对定量,用于筛选用高压氧预处理的缺血耐受大鼠有差异表达的蛋白。蛋白质组学结果经过Western blot和酶联免疫吸附试验进行验证。然后,用小干扰RNA和基因敲除大鼠来进一步确定候选蛋白在高压氧预处理中诱导的内源性神经保护这一关键作用。最后,检测了溶酶体通透性,来阐述这种内在的神经保护作用的机制。

研究人员发现从高压氧预处理获得益处的大鼠血清中有9个蛋白存在差异表达,对这9个蛋白进行筛选和鉴定。Western blot和酶联免疫吸附试验结果显示,胱抑素C和甘露糖结合凝集素蛋白C在高压氧预处理与脑缺血后梗死面积较小的大鼠中发生了特异性改变的蛋白。基因沉默和敲除胱抑素C可以消除高压氧诱导的神经保护作用。此外,高压氧诱导内源性胱抑素C升高可以保护在野生型大鼠卒中后溶酶体膜的完整性,但在胱抑素小干扰RNA输注或敲除鼠中没有这一效应。最重要的是,外源性胱抑素C也可以诱导神经保护作用来对抗缺血/再灌注损伤。

由此可见,胱抑素C是导致内源性神经保护的一个重要因素。这也是一种新的通过维持溶酶体膜完整性的卒中候选治疗手段。
