Cell:“免疫杀死癌细胞”再获重大突破,临床试验效果显著!

登山者和耐力运动员通常会接受高原训练,学习在低氧条件下生存。原来抗癌细胞的免疫系统也可以通过高原训练来提高他们的性能,在《细胞》杂志上发表的一项研究中报道称,来自魏茨曼科学研究所的研究人员表明,免疫系统的杀手T细胞破坏癌变肿瘤的能力在缺氧后更有效。

利用免疫系统对抗恶性肿瘤的方法被称为癌症免疫疗法,在过去的几年中,延长了很多癌症患者的生命,这种方法主要依靠患者血液中的杀手T细胞,在培养皿中帮助它如何识别和摧毁癌细胞,然后再回到病人的血液中。迄今为止,这种方法对某些白血病淋巴瘤效果最好,但对实体肿瘤效果不佳,可能是因为这样的肿瘤与健康器官相比氧气浓度极低,只有0.5%到5%的氧气,而在常规实验室孵化器中,氧气的比例要占20%。

缺氧对肿瘤细胞似乎没什么影响,但是T细胞却很难穿透肿瘤,执行他们杀死的功能。先前的研究表明,T细胞在低氧条件下有助于消灭细胞培养皿中的癌细胞,但他们的实际抗癌能力从来没有测试。

研究人员表示:“杀手T细胞免疫疗法的目标是摧毁癌细胞,但成功率却无法达到百分之百,在此次研究中,通过增加在低氧环境中训练T细胞,我们可以把它们变成更有效的杀手。”

“在细胞免疫疗法,T细胞需要移除到体外进行训练,低氧压力是相对比较容易做到的,这个小小的调整可以显着提高临床治疗的有效性。”
