一项新出炉的研究警告称,即使是失眠一夜,也可能会使大脑充满与老年痴呆症有关的蛋白质块。

研究人员发现,睡眠不足的人一夜之间β-淀粉样蛋白淀粉样蛋白就会显著增加,而这种物质在神经元间会形成妨碍大脑功能的斑块。

该研究作者、美国国家酒精滥用和酒精中毒研究所研究员Shokri Kojori表示,依据此次研究,他认为,即使一个晚上的失眠也会导致大脑充满有害的β-淀粉样蛋白化合物。他说,“我认为这一假设是非常合乎逻辑的,它与先前的研究结论也是一致的。”

研究人员在背景资料中指出,以前的小鼠和人类研究发现,睡眠过少与大脑中淀粉样β蛋白的积累之间存在潜在联系。然而,许多研究依靠的都是自我报告形式的睡眠质量记录。

因此Shokri Kojori和他的团队决定创造一种新的实验方式,能更精确地检测失眠对人类β-淀粉样蛋白水平的影响。

他们招募了20名没有脑部疾病史的健康人,并让他们在实验室里待了两个晚上,一组晚上可以休息,另一组晚上不睡觉。两天后的早晨,参与者接受脑部扫描来评估他们的β-淀粉样蛋白水平。

研究人员发现,与良好睡眠的一组相比,睡眠不足组人员的大脑中β-淀粉样蛋白显著增加。此外,在他们大脑中重要的记忆区域也观察到了β-淀粉样蛋白的增加。

专家认为,每一个神经元都有助于大脑中β-淀粉样蛋白的产生,当人们不睡觉时,他们的神经元会持续工作,可能导致产生更多的β-淀粉样蛋白。

同时,失眠也会导致包括β-淀粉样蛋白在内的大脑废物难以被清除干净,因为在睡眠期间,神经元细胞会缩小,会让大脑废物更容易从脑中被清除。

专家们也指出,还需要做更多的研究,以证明睡眠不足与老年痴呆症的风险增加存在直接联系。

Shokri Kojori指出,未来的一项实验可能会让参与者经历一晚的失眠,然后再好好睡上一晚,然后进行大脑扫描,以证明睡眠是否能恢复大脑健康。

此外,研究人员还需要证明,在大脑中持续高水平的β-淀粉样蛋白,是否会增加神经元之间形成斑块的几率。

该研究发表在最新一期的《美国国家科学院学报》(PNAS)上。
