ARD:体外质谱分析揭示了银屑病关节炎滑液中的骨髓促炎特征

本研究使用质谱流式细胞术(CyTOF)结合转录组学分析来生成匹配的PsA滑液(SF)和血液白细胞的高维数据集,目的是在未受刺激的淋巴和骨髓细胞中体外识别细胞因子的产生。

      目的: 许多免疫细胞群与银屑病关节炎(PsA)的发病机制有关。目前已知PsACD8+T细胞(尤其是产生IL-17的)有关;髓细胞群的作用尚未进行相同程度的探索。本研究使用质谱流式细胞术(CyTOF)结合转录组学分析来生成匹配的PsA滑液(SF)和血液白细胞的高维数据集,目的是在未受刺激的淋巴和骨髓细胞中体外识别细胞因子的产生。

     方法:将新鲜SF和配对血液固定或与蛋白质转运抑制剂一起孵育6 小时。样品用两个 CyTOF面板染色:一个表型面板和一个细胞内面板,包括针对T细胞和骨髓细胞分泌蛋白的抗体。还通过关键扩增细胞群的基因阵列进行了转录组学分析、单细胞RNA-seqELISAPsA SFLEGENDplex分析。

     结果:研究观察到PsA SF的髓样区室相对于血液发生了显著变化,中间单核细胞、巨噬细胞和树突状细胞群出现扩增。在没有任何体外刺激的情况下,经典单核细胞、中间单核细胞和巨噬细胞会自发产生大量的促炎介质骨桥蛋白和CCL2。相比之下,少量自发细胞因子在T细胞中检测到。基因表达分析表明,骨桥蛋白和CCL2的基因在SF中被PsA 单核细胞/巨噬细胞高度上调;并且这两种蛋白质在PsA SF中均升高

     结论:使用多组学分析,该研究生成了PsA SF和血液的综合细胞图谱,以揭示PsA 中具有潜在致病和治疗重要性的关键骨髓促炎模块。骨桥蛋白和CCL2有可能用于PsA诊断和治疗。抑制单核细胞/巨噬细胞激活和功能的疗法也值得研究,这些细胞可能驱动PsA中的炎症。
