CLIN CANCER RES：厚朴酚通过下调存活素增加头颈部鳞状细胞癌放疗敏感性

之前的研究表明存活素在头颈部鳞状细胞癌中多种作用。CLIN CANCER RES近期发表了一篇文章，研究头颈部鳞状细胞癌中存活素表达的临床意义，头颈部鳞状细胞癌细胞进行放疗后存活素在DNA损伤修复中的功能以及厚朴酚通过下调存活素增强放疗效果的能力。

之前的研究表明存活素在头颈部鳞状细胞癌中多种作用。CLIN CANCER RES近期发表了一篇文章，研究头颈部鳞状细胞癌中存活素表达的临床意义，头颈部鳞状细胞癌细胞进行放疗后存活素在DNA损伤修复中的功能以及厚朴酚通过下调存活素增强放疗效果的能力。

作者分析了头颈部鳞状细胞癌患者原发肿瘤组织（n=100）存活素表达情况并与临床特征相联系。使用头颈部鳞状细胞癌细胞系，在体外及体内研究存活素的功能和厚朴酚对存活素表达的影响。研究结果表明，在接受放疗的患者和所有患者中，存活素过表达与淋巴结转移，总生存降低和无病生存降低显着相关。在头颈部鳞状细胞癌细胞中，清除存活素可以增强放疗后DNA损伤和细胞死亡，而存活素过表达则增强细胞存活能力。放疗可以诱导核存活素聚集及其与γ-H2AX和DNA-PKCs之间的分子作用。通过I-Scel核酸内切酶诱导存活素与DNA DSB特异性结合。厚朴酚联合放疗可以显着增强对获得性放疗耐受的头颈部鳞状细胞癌的毒性，在头颈部鳞状细胞癌模型中抑制肿瘤生长。

文章最后认为，存活素是负性预后因素，与放疗导致的DNA损伤修复有关。使用厚朴酚靶向存活素联合放疗是一种有效的治疗措施。
