PNAS:蜈蚣的中药成分可做镇痛剂

蜈蚣是常用的动物中药药材,用于治疗中风和疼痛等疾病。中国科学院昆明动物研究所赖仞研究员领导的研究团队近期对蜈蚣来源的活性多肽及其发挥药效的作用机理进行了较为系统的研究,发现了一系列作用于离子通道的蜈蚣神经毒 (Yang et al, Mol Cell Proteomics. 2012;11:640-50)。

在前期工作基础上,最近该研究团队与澳大利亚Glenn King教授团队合作,从中国金头蜈蚣中识别了一专一的钠离子通道1.7亚型(NaV1.7)抑制剂多肽μ-SLPTX-Ssm6a。NaV1.7离子通道是研发无成瘾性镇痛药物的重要靶标,该通道的选择性抑制剂几乎可用于各种疼痛的治疗。因此,NaV1.7离子通道高度选择性抑制剂是目前镇痛药物研发的热点。金头蜈蚣来源的μ-SLPTX-Ssm6a对离子通道1.7亚型(NaV1.7)具有高度选择性,对其他离子通道没有作用或作用很弱。通过小鼠镇痛动物模型研究表明,μ-SLPTX-Ssm6a具有比吗啡更强的镇痛作用。

最近,该研究成果被美国科学院院刊(Proc Natl Acad Sci U S A)接受发表,论文题目是Discovery of a selective NaV1.7 inhibitor from centipede venom with analgesic efficacy exceeding morphine in rodent pain models (Yang et al, Proc Natl Acad Sci U S A. 2013 Sep 30. [Epub ahead of print]) 。该工作得到了国家自然科学基金委和科技部的支持。
