二代测序技术(NGS)经过优化可以提供可以与之比较的技术表现,尤其是随着唯一标识符(UIDs)的引入。然而,当使用UIDs时,影响数据质量的参数并没有充分的理解。最近,有研究人员利用了一种靶标NGS方法对65个来自膀胱癌病人的血浆样本进行了分析。当使用UIDs时,实验室和生物信息学参数能够影响数据质量。研究人员使用一个225kb的基因板在感兴趣的基因组位置平均得到了249个特异的片段。验证试验确定了38个中的24个变异,这38个变异在一些血浆样本中利用ddPCR鉴定获得。另外,研究人员还利用NGS在相关的肿瘤样本中检测到了4个突变,并没有使用ddPCR技术。从一为膀胱癌患者的连续性血浆样本收集的cfDNA分析表明了与放射成像相比更早的复发检测。最后,研究人员指出,这些发现也许进一步深刻了NGS的技术优势,并且可协调cfDNA分析。
